Abstract:
A high performance liquid chromatographic method was developed for the determination of hydroxysafflor yellow A and salidroside in Jiuwei Shihuihua San, a traditional Chinese medicinal preparation. The samples were analyzed on a Kromasil C
18 column (5 μm, 4.6 mm×250 mm) using acetonitrile-0.1% aqueous phosphoric acid solution (12∶88,V/V) as the mobile phase at a flow rate of 1.0 mL/min, 403 nm as the detection wavelength for hydroxysafflor yellow A and 275 nm for salidroside. Hydroxysafflor yellow A and salidroside showed good relationship at the range of 0.288~180.000 μg/mL and of 4.8~1200.0 μg/mL, respectively. The correlation coefficients of the calibration curve for the two analytes exceeded 0.999. The detection limit was 0.03 μg/mL and 0.5 μg/mL at a signal-to-noise ratio of 3. The recovery of hydroxysafflor yellow A and salidroside was 95.7% ~ 97.4% and 95.9% ~ 98.6%, respectively. The results showed that the method is simple, accurate and repeatable and it is suitable for quality control of Jiuwei Shihuihua San.
