氯酚类物质测定新方法及其与人血清白蛋白的相互作用

New Methods for the Determination of Chlorophenols and the Interaction Between Them with Human Serum Albumin

  • 摘要: 以2,4-DCP为代表,分别考察了溶液酸度、试剂用量、反应温度和反应时间等因素对该体系荧光猝灭强度的影响;由Stern-Volmer方程求出2,4-DCP与HSA的猝灭常数(Kq)及反应热力学参数.新建方法线性范围为0.75~6.50μg/10 mL,检出限为2.26×10-10g/mL(以2,4-DCP计).Kq=3.713×1013L/(mol·s).该方法结合C18柱固相萃取技术,用于水中CPs的测定,相对标准偏差小于2.91%,加标回收率为96.44%~105.94%,结果满意.2,4-DCP对HSA内源荧光的猝灭以静态猝灭为主.

     

    Abstract: Objective: Establishing new method for the determination of chlorophenols(CPs).Studying the association reaction mechanism of CPs with HSA. Methods: The affecting factors on the strength of fluorescence quenching including acidity,dosage,temperature and reaction time etc were investigated.According to Stern-Volmer equation,the quenching constant of the reaction and the basic thermodynamic parameters of the blinding process were obtained. Results: The detection limit was 2.26×10-10 g/mL and the linearity range were 0.75~6.50 μg/10 mL.The fluorescence quenching constant Kq was 3.713×1013 L/(mol·s). Conclusion: When the proposed method was used to determine CPs (2,4-DCP) in water,combined with solid phase extractor(SPE),the satisfactory results were obtained and the relative standard deviation was less than 2.91% and the recovery range was 96.44%~105.94%.Fluorescence quenching mechanism of 2,4-DCP with HSA was a static quenching procedure.
