Abstract:
Traditionally, the internal information of biological tissue samples needs to be obtained by mechanical slicing, which is time-consuming, labor-intensive, easy to cause sample damage and loss of information. The development of tissue clearing and microscopic imaging technology realizes the observation of the internal structure of tissue and even whole body without mechanical slicing. The tissue clearing of lung and kidney of mice were obtained using CUBIC (clear, unobstructed brain imaging cocktails and computational analysis) method, and the imaging effect of the tissue on Olympus spinning-disk confocal microscope and Luxendo light-sheet microscope were compared, which provides a reference for the conditions of lung and kidney tissue clearing and the selection and application of imaging equipment. In addition, tissue expansion and tissue clearing of relatively dense kidney tissue was explored in combination with tissue expansion gel, which provides a reference for high resolution imaging of cleared tissue.
